Figure 4 (A-E)
Illustration of intracellular compartments in a cell and life-cycle of CT Receptor (panel A). Confocal images of 1µM staurosporine treated HGG cell lines at 5 hr immuno-stained with anti-human mouse CT Receptor antibody (mAb1H10), anti-human mouse CTb Receptor antibody (mAb10G6) and anti-human rabbit antibody against intracellular marker Rab11 (panel B) or Rab27a (panel D) primary antibodies followed by secondary antibodies (CT Receptor: anti-mouse IgG2A:AF568; CTb Receptor: anti-mouse IgG1:AF563; Rab11: anti-rabbit:AF488). Images were taken at 20X magnification as Z-stack and are representative of 3 independent experiments. All panels show a single plane selected from the Z-stack. Scale bar represents 100 µm length.
Line graphs in panel D represents change in mean intensity of CT Receptor ( red circles, mAb1H10), CTb Receptor ( purple squares, mAb10G6), Rab11 ( green triangles, recycling vesicles) and Rab27a ( blue inverted triangles, secretory vesicles), and panel E represents colocalisation of CT Receptor (mAb1H10) with Rab11 ( red circles) or Rab27a ( purple squares), and CTb Receptor (mAb10G6) with Rab11 ( green triangles) or Rab27a ( blue inverted triangles) (panel E) respectively, in 1 µM staurosporine treated HGG cell lines over time points 0, 2 and 5 hr. The error bars represent +/- SD from individual experiments (n=2 to 5).
